a-AMINOISOBUTYRIC ACID TRANSPORT IN THE MIDGUT OF TWO LEPIDOPTERAN LARVAE
نویسندگان
چکیده
1. A net absorption of a-aminoisobutyric acid (AIB) takes place in vitro in the midgut of two lepidopteran larvae, Philosamia cynthia Drury and Bombyx mori L. 2. lnP. cynthia the midgut epithelium accumulates AIB from the lumen, while in the same conditions AIB accumulation is not observed in B. mori midgut cells. 3. In P. cynthia, when the lumen is bathed with a low K solution, the net absorption of AIB is reduced and the intracellular accumulation from the lumen is abolished. 4. Brush border membrane vesicles, prepared from the midgut of both species, show a transient K-dependent concentrative uptake of AIB. 5. The relationship between AIB uptake and AIB concentration in the presence of a transmembrane K gradient was studied in B. mori vesicles and the kinetic constants were calculated. 6. These results confirm that there is a K-amino acid co-transport system on the brush border of columnar cells in the midgut of both larvae.

Kinetics of leucine transport in brush border membrane vesicles from lepidopteran larvae midgut.
The kinetics of K(+)-leucine cotransport in the midgut of lepidopteran larvae was investigated using brush border membrane vesicles. Initial rate (3 s) of leucine uptake was determined under experimental conditions similar to those occurring in vivo, i.e. in the presence of delta psi much greater than 0 (inside negative) and a delta pH of 1.4 units (7.4in/8.8out). Leucine and K+ bind to the car...

Magnesium and calcium ions, in concentrations comparable to those reported in the midgut fluids of lepidopteran larvae, bring about the precipitation of most of the tannic acid present in simple solutions buffered at pH 8.0 and 10.0, but not at pH 6.5. In contrast, when tannic acid is added to Manduca sexta midgut fluid, less than 31% of the tannic acid added to the gut fluid is converted to a ...
متن کاملInsight into the Mode of Action of Celangulin V on the Transmembrane Potential of Midgut Cells in Lepidopteran Larvae
Celangulin V (CV) is the main insecticidal constituent of Celastrus angulatus. The V-ATPase H subunit of the midgut cells of lepidopteran larvae is the putative target protein of CV. Here, we compared the effects of CV on the midgut membrane potentials of Mythimna separata and Agrotis ipsilon larvae with those of the Cry1Ab toxin from Bacillus thuringiensis and with those of inactive CV-MIA, a ...
متن کاملA Lepidopteran-Specific Gene Family Encoding Valine-Rich Midgut Proteins
Many lepidopteran larvae are serious agricultural pests due to their feeding activity. Digestion of the plant diet occurs mainly in the midgut and is facilitated by the peritrophic matrix (PM), an extracellular sac-like structure, which lines the midgut epithelium and creates different digestive compartments. The PM is attracting increasing attention to control lepidopteran pests by interfering...
متن کاملEffects of Periplocoside P from Periploca sepium on the Midgut Transmembrane Potential of Mythimna separata Larvae
Periplocoside P (PSP) isolated from the root bark of Periploca sepium contains a pregnane glycoside skeleton and possesses high insecticidal properties. Preliminary studies indicated that PSP disrupts epithelial functions in the midgut of lepidopteran larvae. In the present study, we examined the effects of PSP on the apical and basolateral membrane voltages, Va and Vbl, respectively, of cells ...
